Levodopa, Carbidopa, and Entacapone is a triple-combination antiparkinsonian formulation. Levodopa is the metabolic precursor of dopamine that crosses the blood-brain barrier; carbidopa is a peripheral aromatic L-amino acid decarboxylase (DOPA decarboxylase) inhibitor; and entacapone is a reversible peripheral catechol-O-methyltransferase (COMT) inhibitor. Carbidopa and entacapone block both major peripheral metabolic degradation pathways of levodopa, significantly increasing the plasma half-life and brain delivery of levodopa and smoothing out motor fluctuations. Indicated for Parkinson's disease patients experiencing end-of-dose motor fluctuations ("wearing-off").

Usage Guidelines
Take orally with a full glass of water. Swallow tablets whole; do not break, crush, or chew. Avoid taking with high-protein meals, which compete with levodopa for intestinal and blood-brain barrier amino acid transport. Complete dosing exactly as timed by your neurologist.
General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.
Possible Side Effects
Common: Dyskinesia, nausea, harmless reddish-brown/orange urine discoloration, diarrhea, dizziness, somnolence, orthostatic hypotension. Rare/Severe: Severe dyskinesias, hallucinations, sudden sleep onset episodes while driving, neuroleptic malignant syndrome-like reaction upon abrupt cessation, and rhabdomyolysis.
*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.
Precautions & Warnings
Urine may turn harmless reddish-brown due to entacapone metabolites. Contraindicated with non-selective MAO inhibitors and in patients with history of melanoma or narrow-angle glaucoma. Never stop abruptly to prevent acute hyperpyrexia and rigidity.
