Clinical Overview

Domperidone Tablets (10mg) and Suspension (5mg/5ml) is a peripheral dopamine D2 receptor antagonist antiemetic and prokinetic on the WHO Model List framework. Because it does not readily cross the blood-brain barrier, it acts primarily at the chemoreceptor trigger zone (outside the blood-brain barrier) and gastrointestinal tract, stimulating upper GI motility, increasing LES pressure, and accelerating gastric emptying. Indicated for relief of symptoms of nausea and vomiting of various etiologies, and diabetic gastroparesis.

Usage Guidelines

Take orally 15 to 30 minutes before meals and, if needed, before bedtime. Adults: 10 mg up to 3 times daily (maximum 30 mg/day). Pediatric suspension: Dose strictly based on weight (0.25 mg/kg up to 3 times daily). Treatment duration should not exceed 7 days for acute nausea.

General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.

Possible Side Effects

Common: Dry mouth, headache, abdominal cramps, diarrhea. Rare/Severe: QTc prolongation, ventricular tachycardia, Torsades de Pointes, hyperprolactinemia (galactorrhea, gynecomastia, amenorrhea), and acute dystonic reactions.

*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.

Precautions & Warnings

Contraindicated in patients with moderate or severe hepatic impairment, underlying cardiac conduction disorders, prolonged QTc intervals, heart failure, or prolactinoma. Maximum recommended duration is 7 days.
