Clinical Overview
Tenofovir Alafenamide (TAF) Tablets (25mg) is a targeted prodrug of the nucleotide reverse transcriptase inhibitor tenofovir on the WHO Model List of Essential Medicines. TAF enters hepatocytes through passive diffusion and hepatic OATP1B1/1B3 transporters and is intracellularly hydrolyzed by carboxylesterase 1 (CES1) to tenofovir. This delivers high intracellular antiviral concentrations with over 90% lower systemic plasma tenofovir exposure compared to tenofovir disoproxil fumarate (TDF), dramatically reducing renal tubulopathy and bone mineral density loss. Indicated for the treatment of chronic hepatitis B virus (HBV) infection in adults and adolescents with compensated liver disease.

Usage Guidelines
Take orally 25 mg once daily with food with a glass of water. Swallow whole. High compliance is essential.
General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.
Possible Side Effects
Common: Headache, abdominal pain, fatigue, cough, nausea, back pain. Rare/Severe: Acute exacerbation of hepatitis B following discontinuation, lactic acidosis, and severe hepatomegaly with steatosis.
*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.
Precautions & Warnings
Black Box Warning: Severe acute exacerbation of hepatitis B after discontinuation. Superior renal and bone safety profile compared to TDF. Not recommended in patients with CrCl < 15 mL/min who are not receiving chronic hemodialysis.
