Clinical Overview
Darifenacin Prolonged-Release Tablets (7.5mg) is a potent, selective muscarinic M3 receptor antagonist on the PMBI formulary. It exhibits >9-fold higher selectivity for M3 receptors (responsible for bladder detrusor contraction and salivary flow) over M1 and M2 receptors (responsible for cardiac chronotropy and cognitive function), effectively reducing bladder spasms while sparing cardiac rate and central cognitive faculties. Indicated for the treatment of Overactive Bladder with symptoms of urge urinary incontinence, urgency, and frequency.

Usage Guidelines
Take orally once daily, starting with 7.5 mg once daily with water. Swallow whole; do not chew or crush. Can be taken with or without food. Maximum dose is 7.5 mg daily in moderate hepatic impairment (Child-Pugh B).
General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.
Possible Side Effects
Common: Dry mouth, constipation, dyspepsia, abdominal pain, dry eyes, nausea. Rare/Severe: Urinary retention, severe constipation/bowel impaction, and angioedema.
*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.
Precautions & Warnings
Contraindicated in urinary retention, gastric retention, and uncontrolled narrow-angle glaucoma. Highly advantageous over non-selective anticholinergics in elderly patients due to lack of central M1 receptor blockade (preserves cognitive function).
