Clinical Overview
Diazepam is a prototype long-acting 1,4-benzodiazepine on the WHO Model List of Essential Medicines. It binds to the GABAA receptor complex in the limbic system, thalamus, and hypothalamus, facilitating GABA-mediated inhibitory neurotransmission. Possesses an active desmethyldiazepam metabolite with a prolonged elimination half-life (up to 100 hours). Indicated for management of anxiety disorders, acute alcohol withdrawal syndrome (delirium tremens), skeletal muscle spasm, and status epilepticus.
