Clinical Overview

Deferasirox Dispersible Tablets (180mg, 250mg, 360mg, 400mg, 500mg) is an oral tridentate iron-chelating agent on the WHO Model List of Essential Medicines. It selectively binds ferric iron (Fe3+) in a 2:1 ratio, promoting fecal iron excretion and clearing excessive toxic systemic iron stores without redistributing iron to the brain. Indicated for treatment of chronic iron overload due to frequent blood transfusions in patients with beta-thalassemia major, sickle cell disease, and other chronic anemias (age >=2 years), and non-transfusion-dependent thalassemia syndromes.

Usage Guidelines

Take orally once daily on an EMPTY STOMACH: at least 30 minutes before food with water, at the same time each day. Dispersible tablets: Disperse the tablet completely by stirring in a glass of water, apple juice, or orange juice (100-200 mL) until a fine suspension forms, and drink immediately. Rinse glass with a little juice/water and drink residue. Do NOT chew or swallow tablets whole.

General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.

Possible Side Effects

Common: Nausea, vomiting, diarrhea, abdominal pain, skin rash, mild serum creatinine elevation, transaminase rises. Rare/Severe: Acute renal failure, cytopenias, hepatic failure, gastrointestinal hemorrhage/ulceration, and auditory/visual disturbances (cataracts, hearing loss).

*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.

Precautions & Warnings

Black Box Warning: Renal failure, hepatic failure, and gastrointestinal hemorrhage (can be fatal); monitor serum creatinine and creatinine clearance in duplicate prior to initiation and monthly thereafter. Monitor serum ferritin monthly. Perform baseline and annual auditory and ophthalmologic testing.
