Clinical Overview

Progesterone (Injection IP 100mg/2ml) is a bioidentical progestogen on the WHO Model List of Essential Medicines. Identical to human ovarian luteal progesterone, it transforms the proliferative endometrium into a secretory endometrium, promotes blastocyst implantation, suppresses myometrial oxytocin receptor expression, and inhibits uterine contractions to maintain pregnancy. Indicated for luteal phase support in assisted reproduction (IVF/ICSI), threatened and recurrent miscarriage, prevention of preterm birth in women with short cervix, secondary amenorrhea, and endometrial protection during estrogen HRT.

Usage Guidelines

For Deep Intramuscular (IM) injection ONLY: 50 mg to 100 mg deep IM daily or on alternate days as prescribed by the reproductive endocrinologist/obstetrician. Warm vial slightly between palms to decrease viscosity of oily vehicle; inject slowly using a deep 21G/22G needle. Continue through 10-12 weeks of gestation as directed.

General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.

Possible Side Effects

Common: Drowsiness/somnolence (oral route), dizziness, headache, breast tenderness, abdominal bloating, nausea, and vaginal discharge/leakage (vaginal route). Rare/Severe: Thromboembolic events (DVT, PE), cholestatic jaundice, depression, and severe allergic reactions.

*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.

Precautions & Warnings

Use caution when driving or operating machinery due to central sedative metabolites (allopregnanolone) when taken orally. Contraindicated in active venous thromboembolism, severe hepatic impairment, undiagnosed vaginal bleeding, porphyria, and missed abortion. Safe and indicated during early pregnancy under specialist care.
