Clinical Overview
Clozapine is a dibenzodiazepine prototype atypical antipsychotic on the WHO Model List of Essential Medicines. It possesses weak affinity for dopamine D2 receptors and potent blockade of D4, 5-HT2A, alpha-1, and muscarinic receptors. Demonstrates unique superiority in reducing suicidal behavior and treating refractory psychosis without producing extrapyramidal symptoms or tardive dyskinesia. Strictly reserved for treatment-resistant schizophrenia and reducing suicidal behavior in schizophrenia or schizoaffective disorder.
