Clinical Overview
Buspirone Hydrochloride is an azaspirodecanedione anxiolytic agent that acts as a high-affinity partial agonist at presynaptic and postsynaptic serotonin 5-HT1A receptors, with moderate affinity for dopamine D2 autoreceptors. Unlike benzodiazepines, it possesses zero affinity for GABAA receptors, producing effective sustained anxiolysis without causing sedation, cognitive impairment, muscle relaxation, anticonvulsant activity, or physical dependence and withdrawal. Indicated for generalized anxiety disorder (GAD) and short-term relief of anxiety symptoms.

Usage Guidelines
Take orally with water, divided twice or thrice daily. Take consistently either always with food or always without food, as food substantially increases bioavailability. Requires 2 to 4 weeks of continuous daily therapy to achieve full therapeutic anxiolysis. Not effective for acute PRN panic relief.
General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.
Possible Side Effects
Common: Dizziness, drowsiness, headache, nausea, nervousness, lightheadedness, excitement. Rare/Severe: Serotonin syndrome (when combined with MAOIs or SSRIs), akathisia, and chest pain.
*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.
Precautions & Warnings
Does not possess cross-tolerance with benzodiazepines; if switching from a benzodiazepine, the benzodiazepine must be tapered gradually. Contraindicated with MAO inhibitors (risk of severe blood pressure elevation). Avoid large quantities of grapefruit juice.
