Panitumumab Infusion (100mg, 400mg) is a recombinant, fully human IgG2 monoclonal antibody on the WHO Model List of Essential Medicines that binds specifically to the human Epidermal Growth Factor Receptor (EGFR / HER1). It competitively prevents binding of endogenous EGF and TGF-alpha, halting EGFR dimerization, receptor autophosphorylation, and downstream pro-proliferative intracellular cascades. Indicated for wild-type KRAS and NRAS (exon 2, 3, and 4) metastatic colorectal carcinoma (mCRC), as first-line therapy with FOLFOX, or as monotherapy following progression on oxaliplatin, irinotecan, and fluoropyrimidines.

Precautions & Warnings
Black Box Warning: Dermatologic and soft tissue toxicity (severe acneiform rash occurs in >90% of patients; withhold or reduce dose for grade 3+ toxicity); apply prophylactic moisturizers and sunscreens. Black Box Warning: Ineffective in patients with mutated RAS (KRAS/NRAS) colorectal tumors; genetic testing for wild-type RAS is mandatory prior to therapy. Monitor serum magnesium and calcium weekly.
