Clinical Overview
Dasatinib Tablets (20mg, 50mg, 70mg) is a second-generation dual BCR-ABL and SRC family tyrosine kinase inhibitor on the WHO Model List of Essential Medicines. It binds to both the active and inactive conformations of the BCR-ABL kinase domain with >300-fold higher potency than imatinib, overcoming most imatinib-resistant BCR-ABL kinase domain mutations (except the gatekeeper T315I mutation). Indicated for newly diagnosed Ph+ CML in chronic phase, and accelerated/blast phase CML or Ph+ ALL with resistance or intolerance to prior therapy including imatinib.

Usage Guidelines
Take orally once daily (100 mg once daily for chronic phase; 140 mg once daily for advanced phase/blast crisis) with or without food. Swallow tablets whole; do NOT crush, split, or chew. Avoid taking antacids within 2 hours of dasatinib (gastric acid suppression reduces bioavailability).
General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.
Possible Side Effects
Common: Pleural effusion (occurs in up to 15-30% of patients), dyspnea, fluid retention, diarrhea, headache, musculoskeletal pain, fatigue. Rare/Severe: Severe myelosuppression, pulmonary arterial hypertension (PAH), severe gastrointestinal hemorrhage, and QT interval prolongation.
*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.
Precautions & Warnings
Black Box Warning: Myelosuppression (severe neutropenia, thrombocytopenia) and bleeding events. Monitor for dry cough, dyspnea, and pleuritic chest pain; pleural effusion often requires temporary drug interruption and diuretic/corticosteroid therapy. Screen for pulmonary arterial hypertension if persistent dyspnea develops. Avoid H2 blockers and PPIs.
