Conjugated Estrogens 0.625 mg Tablets is a mixture of natural estrogenic substances (principally sodium estrone sulfate and sodium equilin sulfate) on the WHO Model List of Essential Medicines. It binds to nuclear estrogen receptors (ER-alpha and ER-beta), activating gene transcription to replenish declining endogenous estrogen levels in postmenopausal women. Indicated for the management of moderate to severe vasomotor symptoms (hot flashes, night sweats) and vulvovaginal atrophy associated with menopause, and prevention of postmenopausal osteoporosis.

Usage Guidelines
Take orally 0.625 mg once daily with water, with or without food. Administer at the lowest effective dose for the shortest duration consistent with treatment goals. In women with an intact uterus, an appropriate progestogen MUST be co-prescribed for 12-14 days each month to protect against endometrial hyperplasia and carcinoma.
General Administration Advice: Adhere strictly to the prescribed dosage, frequency, and treatment course. For oral medications, swallow with sufficient water. Never stop antibiotics, psychiatric medications, or antihypertensives abruptly without clinical direction.
Possible Side Effects
Common: Breast tenderness, nausea, headache, fluid retention/edema, leg cramps, breakthrough bleeding, and abdominal bloating. Rare/Severe: Endometrial cancer (if unopposed by progestin), breast cancer, venous thromboembolism (DVT/PE), stroke, myocardial infarction, and gallbladder disease.
*Note: Individual physiological responses vary. While many patients experience few or no side effects, notify your treating physician if adverse reactions persist or if hypersensitivity (angioedema, skin rash, dyspnea) occurs.
Precautions & Warnings
Black Box Warning: Unopposed estrogens in women with a uterus increase the risk of endometrial cancer; increased risk of stroke, DVT, pulmonary embolism, and breast cancer (WHI study). Contraindicated in undiagnosed abnormal genital bleeding, known or suspected breast or estrogen-dependent cancer, active DVT/PE, arterial thromboembolic disease, and severe liver dysfunction.
